STARCH SYNTHASE 4 is required for normal starch granule initiation in amyloplasts of wheat endosperm

چکیده

Starch granule initiation is poorly understood at the molecular level. The glucosyltransferase, STARCH SYNTHASE 4 (SS4), plays a central role in Arabidopsis leaves, but its function cereal endosperms unknown. We investigated of SS4 wheat, which has distinct spatiotemporal pattern during grain development. generated TILLING mutants tetraploid wheat (Triticum turgidum) that are defective both homoeologs. morphology endosperm starch was examined developing and mature grains. deficiency led to severe alterations morphology. During early development, while wild-type initiated single ‘A-type’ granules per amyloplast, most amyloplasts mutant formed compound due multiple initiations. This phenotype similar deficient B-GRANULE CONTENT 1 (BGC1). also reduced content leaves pollen propose BGC1 required for proper control development leads A-type amyloplast. absence either protein results variable number initiations amyloplast formation.

Heat inactivation of starch synthase in wheat endosperm tissue.

The effect of temperature on accumulation of starch was studied in grain slices of wheat (Triticum aestivum cv SUN9E), taken 15 days after anthesis. As compared with pretreatment of such slices at 25 degrees C, pretreatment at 30 or 35 degrees C reduced the subsequent conversion of sucrose to starch. In contrast to rice (Oryza sativa cv Calrose), pretreatment of wheat soluble starch synthase in...

PROTEIN TARGETING TO STARCH Is Required for Localising GRANULE-BOUND STARCH SYNTHASE to Starch Granules and for Normal Amylose Synthesis in Arabidopsis

The domestication of starch crops underpinned the development of human civilisation, yet we still do not fully understand how plants make starch. Starch is composed of glucose polymers that are branched (amylopectin) or linear (amylose). The amount of amylose strongly influences the physico-chemical behaviour of starchy foods during cooking and of starch mixtures in non-food manufacturing proce...

Degradation of Glucan Primers in the Absence of Starch Synthase 4 Disrupts Starch Granule Initiation in Arabidopsis*

Arabidopsis leaf chloroplasts typically contain five to seven semicrystalline starch granules. It is not understood how the synthesis of each granule is initiated or how starch granule number is determined within each chloroplast. An Arabidopsis mutant lacking the glucosyl-transferase, STARCH SYNTHASE 4 (SS4) is impaired in its ability to initiate starch granules; its chloroplasts rarely contai...

Suppression of starch synthase I expression affects the granule morphology and granule size and fine structure of starch in wheat endosperm

Studies in Arabidopsis and rice suggest that manipulation of starch synthase I (SSI) expression in wheat may lead to the production of wheat grains with novel starch structure and properties. This work describes the suppression of SSI expression in wheat grains using RNAi technology, which leads to a low level of enzymatic activity for SSI in the developing endosperm, and a low abundance of SSI...

Blue maize: morphology and starch synthase characterization of starch granule.

The use of pigmented maize varieties has increased due to their high anthocyanins content, but very few studies are reported about the starch properties of these grains. The aim of this work was to isolate the starch granules from pigmented blue maize and carry out the morphological, physicochemical, and biochemical characterization studies. The proximate composition of starch granules showed h...
